Lets just say by some strange happenings the trench (6 mile deep trench) suddenly filled with lava or dirt in just a few seconds what would happen to the displaced water?

Actually, that sort of thing happens every now and then. The side of a mountain on the west coast of South America fell off and plunged into the Pacific ocean, causing a wave that traveled all the way to Japan and washed out a few coastal towns there. That was only a few hundred feet of rock. Six miles would be what we call "biblical".
